Pcra helicase

Splet12. jun. 2015 · In this study we show that without the essential Bacillus subtilis accessory DNA helicase, PcrA, the replication machinery slows down at certain regions of the chromosome in a transcription-dependent manner. PcrA is essential to life but incomplete depletion of PcrA only partially inhibits cell survival.

Splet01. mar. 1999 · Whereas L3 stimulates the PcrA helicase activity by promoting co-operative PcrA binding onto its DNA substrate, RepD stimulates the PcrA helicase activity by increasing the processivity of the enzyme and enables PcrA to displace DNA from a nicked substrate. The implication of these results is that PcrA is the helicase recruited into the ... Splet09. jan. 2024 · DNA helicase activity of PcrA is not required for displacement of RecA protein from DNA or inhibition of RecA-mediated DNA strand exchange. Helicases are classified in 6 groups (superfamilies) based on their shared sequence motifs. Helicases not forming a ring structure are in superfamilies 1 and 2, and ring-forming helicases form part of superfamilies 3 to 6. Helicases are also classified as α or β depending on if they work with single or double-strand DNA; α helicases work with single-strand DNA and β helicases work with double-strand DNA. They are also classified by translocation polarity. If translocation occur… Splet02. apr. 1999 · We have determined two different structures of PcrA DNA helicase complexed with the same single strand tailed DNA duplex, providing snapshots of … SpletBy means of in silico mutation studies, we identified the residue Q254 as being crucial for the coupling of ssDNA translocation to the actual catalytic event. An emerging body of evidence suggests that their different cellular functions are directed by interactions with partner proteins that target unwinding activity to appropriate substrates.

Detailed kinetics of PcrA activity were measured using … Splet01. sep. 2006 · PcrA is an ATP-driven 3′ to 5′ helicase responsible for the unwinding of double-stranded DNA (dsDNA) (Velankar et al., 1999, Soultanas et al., 2000) and is the …

SpletThe helicase activity of these two proteins is also enhanced by additional proteins, such as MutL and the ϕΧ174 bacteriophage cisA protein in the cases of UvrD and Rep, respectively ( 7–9). PcrA is an essential helicase in many bacteria with a role in genomic DNA repair and in asymmetric rolling-circle plasmid replication ( 10). Splet31. jan. 2012 · PcrA Biochemistry. SpletThe helicase can then unwind complete plasmids of several thousand bases pairs, although a single PcrA on its own is unable to unwind more than a few base pairs. DNA polymerases have also been shown to activate helicase activity, and many helicases will only work processively as part of larger complexes (Donmez and Patel 2006 ). SpletAbstract Recently determined crystal structures of PcrA helicase complexed with a DNA substrate have revealed details of the helicase mechanism. PcrA and UvrD helicases have been shown to be functional as monomers, challenging previous suggestions that all helicases are required to be oligomeric.

Splet17. jul. 2001 · In this model, PcrA helicase is functional as a monomer that separates duplex DNA in a processive manner by coupling two distinct activities.
